Measuring Cell Metabolism - Transient Micro-chamber Video

Agilent Seahorse XF analyzers utilize a patented transient microchamber, which enables sensitive, precise, and nondestructive metabolic measurements in real time.
Seahorse XF analyzers make use of a disposable sensor cartridge. The sensor contains two fluorophores, one for oxygen (O2) and the other for pH. The measurements are accomplished by the instrument sensor cartridge being lowered to 200 microns above the cells, creating the transient microchamber. Changes in oxygen concentration and pH are automatically calculated and reported as Oxygen Consumption Rate (OCR) and ExtraCellular Acidification Rate (ECAR) in real time, enabling time-resolved, kinetic data of cell metabolism.
The sensors that enable measurement of mitochondrial respiration and glycolysis are built into the sensor cartridge- no dyes or exogenous labels are required. Sensor cartridges are sold as part of a FluxPak and are specific for instrument type.
