Applications

Intact Protein Characterization

LC/MS is widely used by the biopharmaceutical industry for therapeutic protein characterization. The accurate mass provided by time-of-flight (TOF) LC/MS allows for confirmation of the synthetic protein, its impurities, and post-translational modifications (PTM). It can also be used for calculation of drug-to-antibody ratios (DAR) for antibody drug conjugates (ADC), relative quantitation, glycoform, and subunit analyses. Coupled to the Infinity III Bio LC, this is a premier system for sensitive, high-resolution analysis of intact proteins.

Oligonucleotide Quantification and Impurity Analysis

Oligonucleotides have a wide range of applications in genetic testing, research, and forensics. Synthesis of therapeutic oligonucleotides often results in a complex mixture of oligonucleotides and their impurities. Liquid chromatography with high-resolution mass spectrometry allows for profiling of the mixture, relative quantification of the products, and identification by accurate mass. The 6230 LC/TOF is ideal for oligonucleotide analysis, including quantification of synthetic oligos, and impurity analysis as low as 0.5% of the main compound.

Released Glycan Analysis

Critical Quality Attributes (CQA), such as glycosylation, have a significant impact on the efficacy and stability of therapeutic proteins. Glycosylation has historically been analyzed by fluorescence labeling, which does not have sufficient specificity for more complex sample types. The 6230 LC/TOF offers the accurate mass and high resolution required for comprehensive characterization of these glycoforms. This technology can easily be deployed for glycan profiling in the development, manufacturing, and monitoring of biotherapeutics.
