General information

To ensure kit specifications are met, we are constantly evaluating our reagent kits for the Agilent Fragment Analyzer systems. We recently evaluated the injection methods for three qualitative kits: 

  • Agilent dsDNA 910 Reagent kit (DNF-910)
  • Agilent dsDNA 915 Reagent kit (DNF-915)
  • Agilent dsDNA 930 Reagent kit (DNF-930)

During this evaluation, we noticed the occurrence of sample overloading. To reduce saturation in the higher concentration range, the injection voltage was lowered to optimize the signal. With the new lower injection method, the signal was reduced two-fold over the concentration range without affecting kit specifications.

To differentiate the method files, the new method file will display the injection voltage at the end of the method name. Please refer to the table for a list of kits and the affected methods.

Technical information

The examples below show the old (green line) and new (blue line) injection conditions and the overall improvement through the concentration range. An improved linear progression through the higher concentration ranges can be seen.

Software information

Update your injection methods of your Agilent Fragment Analyzer system by following one of the two options: 
Download the new methods and add to your PC (option 1), or manually change the voltage injection/default settings of the methods on your PC and rename the methods (option 2).

Option 1:

The following steps are required to add the new injection methods to the correct PC location:

  1. Select the method file based on the system you own, to download:
    - Download 5200/5300 method files
    - Download 5400 method files
  2. Unzip the method files 
  3. Select/copy the desired new method file to be added to the Fragment Analyzer Controller software

There will be 3 folders as follow: 22 cm, 33 cm, and 55 cm 

  1. Go to computer C: drive > Agilent Technologies > Methods 
  2. Select the matching folder: 22 cm, 33 cm, or 55 cm
  3. Add/paste the new downloaded method file to the appropriate folder. 
  4. Restart the PC for changes to take effect.

Option 2: 

To manually change the method/injections voltage and rename the current method, please follow the steps listed with the enclosed pdf document, available here.
