Proteins carry out a wide variety of molecular functions, due in part to their ability to fold into almost unlimited conformations. In return, their effectiveness in performing these functions is highly dependent on the protein’s structure. It is therefore key for life science researchers and developers of new protein-based therapeutics to have a good understanding of the protein’s structure and structural changes. Technologies that were traditionally used for protein structure analysis include: Circular dichroism (CD) X-ray crystallography Nuclear magnetic resonance (NMR) spectroscopy These techniques usually produce highly accurate results but suffer from sampling and operational challenges: Complex and time-consuming sample preparation Limited to certain sample forms (e.g., crystalline proteins) Requires highly trained operators In a recent application note, researchers at the Agilent Global Solution Development Center (GSDC) in Singapore demonstrated how the Agilent Cary 630 FTIR spectrometer with the Agilent MicroLab software can be used to quickly and easily estimate the secondary structure of proteins in solution. Protein Secondary Structure Estimation Using the Agilent Cary 630 FTIR Spectrometer (5994-7808EN) The sample measurements were completed in a few minutes, and the data analysis was straightforward—only taking a few minutes, without requiring highly skilled technical staff. This methodology can complement traditional technologies as it opens the way to fast secondary structure analysis for: rapid quality check of protein samples, comparative studies between similar proteins, comparative studies between protein therapeutics and their biosimilars, quickly assessing the impact of post-translational modifications on structure, correlating structural changes with functional changes observed with other analytical techniques.
