Enzyme Source
  • Recombinant gene from Elizabethkingia meningoseptica, expressed in E. coli. The source organism was previously known as Chryseobacterium [Flavobacterium] meningosepticum. Enzyme is also known as Endoglycosidase F2, endo-beta-N-acetylglucosaminidase.
Enzyme Specificity
  • The enzyme cleaves within the di-N-acetyl chitobiose (GlcNAc-GlcNAc) core of high mannose and biantennary complex type N-linked glycans of oligosaccharide, glycopeptide, or glycoprotein substrates. A single GlcNAc residue remains attached to the molecule after hydrolysis, the rest of the glycan is released. This specificity is distinct from PNGase F (GKE-5006), which cleaves between asparagine and the innermost GlcNAc of N-linked glycans. EndoF2 cleaves between GlcNAc-GlcNac in biantennary glycans at a rate approximately 20 times higher than high mannose glycans. The enzyme is not active towards hybrid type or tri- and tetraantennary oligosaccharides.

  • One milliunit is defined as the amount of enzyme that will hydrolyze 1 nM of the dansylated porcine fibrinogen glycopeptide substrate DnsVGEN(CHO)R per minute at pH 4.5 and 37°C.
