Utilizing High Throughput AssayMAP Bravo Platform for Assessing Drug Stabilities, Pharmacokinetics and Immunogenicity in Drug Development Environments

High-throughput and automated sample preparation techniques are essential for achieving accurate and precise analyses in drug development. In our laboratory, the AssayMAP Bravo platform has been employed for a range of applications, including in vitro serum stability (IVSS) assessments, absolute quantitation for bioanalysis, pharmacokinetic (PK) studies, and immunogenicity assessments via Immunopeptidomics. For IVSS assay, we use AssayMAP Bravo for automated purification of biologics (e.g., antibody, antibody drug-conjugates (ADCs)) from serum using biotinylated capture reagents and streptavidin cartridges. We also utilize AssayMAP Bravo for the purification of drugs following digestion for absolute quantitation and pharmacokinetics assessment. For these applications, either two-step immobilization followed by affinity purification or direct affinity enrichment with protein A cartridge are being used depending on the project goal.
