We are using it with Chromeleon. My main question is: Is it possible to do a 3D (3D Data Field) recording with my FLD setup? The 2 Pictures are my FLD Method if there are any blunt errors. I would be thankfull if someone could point them out.

Hi there. With a DAD, a signal was collected at 254 nm but I would like to analyze it at 260 nm. The detector was set to store the signal at 254 nm only, but since it was also set to store spectra, I'm hoping to find a workaround in spite of the detector being set to store at a single wavelength.
How to carry out 2D (monolayer) vs 3D (Tumerosphere) Real time ATP rate assay in Agilent XFe 24 seahorse instrument ?
How to carry out 2D (monolayer) vs 3D (Tumerosphere) Real time ATP rate assay in Agilent XFe 24 seahorse instrument ?

Hi I have grown 3D tumerosphere of my cancer cell line I need to see the metabolic difference with respect to 2D monolayer by seahorse ATP rate assay. The instrument that is currently available at our facility is Ailent XFe 24. Just before the assay can I trypsinize the 3D cultured cells for seeding and make them adhere to the poly D lysine coated plate for the assay as we do for suspension cultures ?

For a more detailed explanation, I am following this method: Extracting Specific Absorbance Signal from 3D IsoPlot with OpenLab ChemStation: https://community.agilent.com/knowledge/chromatography-software-portal/kmp/chromatography-software-articles/kp764.extracting-specific-absorbance-signal-from-3d-isoplot-with-openlab-chemstation then this: How to Export XY Data from Chemstation or OpenLab ChemStation to a CSV File Then combining the saves into one file with Time and Absorbance values for each wavelength. e.g Time 400 410 420 0.00145 1.173496 1.142502 1.424313 0.008117 1.178265 1.146317 1.426697 I found this which is similar however is for the online/current runs only and not for already saved runs, and also for pre-set wavelengths and not spectrum: RE: Post-run Macro to export Signal Raw Data to ASCII I have been having troubles trying to adapt this macro to be used in my case.
