However, I was able to see our internal standard (23:0). With the heat issue and trying to move away from DCM we tried a few new methods. Which each new method (listed below) we were able to see the FAME in samples (salmon SRM and olive oil) as expected so that was an improvement, but we were no longer able to see our internal standard. Based on our ability to see similar sized FAME I do not believe it is an instrument issue, but I am not able to find anything about why our internal standard is not showing up. For preparation of the internal standard we dissolved 20.7 mg 23:0 into 100 mL of hexane, but I have a feeling more needs to be done but I have no idea what. I’ve attached the chromatograms for the internal only (70C Jenkins FAME IS Only) and one where the internal did not show up but other FAME did (Salmon Fame Meth 2) along with the latest tune report and the GC-MS method we are using. It is an Agilent 8890 GC/5977B MS with a HP-5MS UI column. While we do have a FATWAX column I regularly run analyses for neutral lipids and the GC is used by other so I would prefer not to regularly change the column. Method 1 (suggestion from Paul Salverda) Add 80 uL of 23:0 standard to tube and evaporate with nitrogen Add 5 mL hexane, vortex Add 200 uL 2M KOH in MeOH, vortex Take top layer, evaporate with nitrogen Method 2 (Agilent application note for milk) Add 80 uL of 23:0 standard to tube and evaporate with nutrogen Add 10 mL hexane, 1 mL sodium methoxide, vortex 1 min Take top layer, evaporate with nitrogen Method 3 (Jenkins, 2010) 100 uL 23:0 standard, evaporate under nitrogen Add 2 mL sodium methoxide (0.5M), vortex Incubate at 50C for 10 min, cool 5 min Add 3 mL 5% methanolic HCl, vortex Incubate at 80C (we tried multiple temperatures) for 10 min, cool 7 min Any suggestions/insights would be greatly appreciated.
