Biotherapeutics such as monoclonal antibodies (mAbs) represent one of the fastest growing classes of protein-based drugs in the rapidly growing biopharmaceutical industry. Therapeutic proteins are first digested by proteases and then analyzed using LC/MS/MS techniques. Optimized and reliable sample preparation is the key first step toward reproducible peptide maps with minimum artifacts introduced in the process. Ultra-high performance LC and reversed phase chemistries enable high resolution and reproducible separation for the digests. Sensitive and accurate Q-TOF mass spectrometry provides confirmation of peptide sequences and PTM identification, including disulfide maps and location of PTMs.
