Proteinase K is a broad spectrum protease that is widely used in DNA purification. It is supplied as a lyophilized powder with a minimum specific activity of 30 U/mg at a reaction temperature of 35°C.
Unit Definition: One unit will hydrolyze casein to produce the absorbance equivalent to 1 µM of tyrosine/minute at pH 7.5, 37°C
Reaction Conditions: 50 mM Tris-HCl (pH 7.5), 5 mM CaCl2, 1 µg DNA and 1 unit enzyme in a 25 µL volume. Incubate 30 minutes at 37°C
Storage Conditions: Store lyophilized or in 10 mM CaCl2 at 20°C
