RTCA Software Pro

RTCA Pro software icon

RTCA Software Pro for xCELLigence RTCA DP, SP, MP, HT, and S16 has four modules, Basic, Immunotherapy, Compliance and Virology.

  • The xCELLigence RTCA Software Pro is an integrated software package for running and analyzing real time cell analysis data from xCELLigence RTCA DP, SP, MP, HT, and S16 instruments. Two modules are available: RTCA Basic and Immunotherapy. In addition, the Compliance and Virology modules can be added on.
  • The basic module offers algorithms such as slope, cell index doubling time, % of control, and EC50/ IC50 calculations. The immunotherapy (IMT) module is designed to measure cytolysis of target tumor cells by effector cells and other molecules, includes experimental templates for immunotherapy studies, and automatically calculates effector to target cell (E:T) ratios, % cytolysis.
  • The virology module is an add-on module for either RTCA basic and RTCA IMT. It streamlines traditional viral titer methods, including TCID50 and VRNT, by monitoring viral cytopathic effects (CPE) over time with customizable calculation methods and user-defined CPE thresholds.
  • The compliance support mode is an add-on module for either RTCA basic and RTCA IMT. When the compliance support mode is activated, RTCA Software Pro supports users in achieving requirements of FDA 21 CFR Part 11 to ensure the authenticity and integrity of electronic data.
  • Easy-to-use and intuitive xCELLigence RTCA Software Pro analyzes real-time data, generates diverse plot types and automatically calculates dose response curves and parameters such as % cytolysis, IC50 or KT50
  • Quickly evaluate efficacy of different treatments with automated normalization and compensation features
  • Calculate slope, Cell Index doubling time, % of control, and EC50/ IC50 with RTCA Pro Basic
  • Evaluate the potency of immune cells (CAR-T, Natural Killer, Tumor-infiltrating lymphocytes), checkpoint inhibitors, antibody dependent cell-mediated cytotoxicity (ADCC) with RTCA Software Pro IMT (Immunotherapy)
  • Determine viral titer and neutralizing activity (TCID50 and VRNT) using customizable formulas and user-defined thresholds with the add-on Virology module
  • Support for FDA 21 CFR Part 11 compliance to ensure the authenticity and integrity of electronic data

Overview

How it works

Simple assay setup

RTCA Software Pro allows users to conveniently set up and analyze experiments. The plate window handles all functionalities relevant to a single E-Plate. Simply enter the cell name, number, treatment name, and concentration for each well using the intuitive software interface. The color-coded plate map provides a visual reference to the conditions being tested. Easily load a template file to create a new experiment, save a current experiment template, and export experiment details as Excel, text, or PDF files.

Streamlined real-time data acquisition and analysis

With RTCA Software Pro, one interface is used for both real-time data acquisition and analysis. To obtain early insights, users can visualize and analyze acquired data while an experiment is ongoing. The Analysis Method feature allows users to save time by loading saved analysis parameters to the Cell Index, Well Graph, and Data Analysis pages. Select and simultaneously display relevant data from specific wells, generate dose response curves, calculate IC50/EC50 values, and create customized reports and publication quality figures.

RTCA software license selection guide

RTCA Software Pro is an integrated software package for running and analyzing real-time cell analysis data from xCELLigence RTCA S16 DP, SP, MP and HT instruments. The RTCA Basic license offers algorithms such as slope, cell index doubling time, % of control, and EC50/IC50 calculations. The IMT license provides powerful immunotherapy analysis tools. The virology license streamlines the calculation of TCID50 and VRNT. The compliance license supports FDA 21 CFR Part 11 compliance in a GMP environment.

Applications

Immune cell killing assays

Typical immune cell killing assays are labor-intensive and require users to collect multiple timepoints using a combination of techniques. In contrast, the label-free xCELLigence RTCA instruments provide a complete view of immune cell killing and closely mimics activity in vivo. Obtain highly sensitive, reproducible, and direct measurements of immune cell killing potency in real time. This simple and high throughput workflow also provides greater insight into cellular mechanism of action, serial killing, and exhaustion.

Viral cytopathic effect assays

Advance your anti-viral therapy or vaccine development by accurately measuring viral cytopathic effects (CPE) in real time with Agilent xCELLigence RTCA instruments, without the use of labor-intensive plaque assays. Monitor viral CPE automatically, from minutes to days, and greatly reduce workload and manual handling of samples. Measure cell proliferation kinetics, identify the optimal time point for viral infection with different cell seeding densities, and assess virus-mediated cytopathogenicity in one workflow.

Cytotoxicity assays

Determine cytotoxic effects of drugs or compounds by monitoring cell proliferation, cell size or morphology, and cell-substrate attachment quality in real time. Monitor the kinetics of long-term cellular responses with this simple and high-throughput cytotoxicity assay. Make informed decisions about the timing of cell treatments and easily generate dose-response curves at multiple time points, without increasing the workload.

Technology

Cellular impedance explained

The functional unit of a cellular impedance assay is a set of gold microelectrodes fused to the bottom surface of a microtiter plate well. When submersed in an electrically conductive solution (such as buffer or standard tissue culture medium), the application of an electric potential across these electrodes causes electrons to exit the negative terminal, pass through bulk solution, and then deposit onto the positive terminal to complete the circuit.
