Smarter live-cell imaging for faster, more confident decisions

 • The Agilent xCELLigence RTCA eSight software drives the Agilent xCELLigence RTCA eSight instrument with flexible assay set up, real-time impedance and imaging acquisition, and integrated analysis tools. The RTCA eSight supports a broad range of labware—including the Agilent E-Plate VIEW 96, standard 96-well plates, and formats from 6- to 384-well plates, dishes, and flasks. It offers both real-time and offline modes for continuous monitoring or post-run analysis. With version 1.4.0, the software introduced support for up to five 96-well regular imaging microplates, 3D organoid workflows, and features to support 21 CFR Part 11 compliance and GMP environments.
 • The base software provides the core functionality to set up, run, and analyze real-time cell assays. By combining impedance and imaging, it monitors adhesion, proliferation, viability, and cytotoxicity in 2D cultures. Built-in tools such as slope, doubling time, percent of control, and EC50/IC50 calculations ensure reliable, reproducible results.
 • With version 1.5.0, an optional AI add-on license introduces AI-powered, label-free cell segmentation for automated cell counting, faster analysis, and more reproducible image analysis workflows.
 • Additional application modules expand these capabilities with immunotherapy assays, 3D spheroid and organoid analysis, single-cell tracking, dilution cloning, and wound healing. This modular design gives researchers the flexibility to study processes ranging from immune cell killing to migration and clonal development.
 • Capture more from a single setup with dual impedance and imaging readouts, multimodal data acquisition, and powerful analysis tools for information-rich results.
 • Monitor and quantify cell health, proliferation, cytotoxicity, and apoptosis in real time using simultaneous impedance and imaging.
 • Evaluate and compare immunotherapies, CAR-T, NK cells, and antibody treatments with automated immune cell killing and potency assays.
 • Expand throughput and accelerate discovery by running up to 5x96 imaging plates simultaneously without hardware changes.
 • Model and analyze complex biology with advanced 3D workflows, including spheroid growth, invasion, and patient-derived organoid studies.
 • Track and characterize single-cell behavior, clonal populations, and wound healing dynamics through automated cell-by-cell analysis.
 • Ensure data integrity and regulatory compliance with built-in 21 CFR Part 11 features like electronic signatures, audit trails, and GMP support
 • Reduce image analysis time using AI-driven, label-free cell segmentation for automated cell counting with a one-click workflow

Overview

How it works

AI-powered brightfield cell quantification

Apply label-free AI segmentation to brightfield images to accurately identify and count cells across time. Generate quantitative growth curves directly from imaging data in standard 2D workflows—no staining required.

Deeper insights with dual-mode data collection and multiplexing

The RTCA eSight Software streamlines simultaneous real-time capture and acquisition of dual impedance and live-cell imaging data, all in the same well. This unique dual-readout approach provides continuous kinetic monitoring without disturbing your cultures, ensuring physiologically relevant results.

Track proliferation curves, cytotoxicity readouts, or migration profiles while experiments are running, and apply saved analysis parameters for consistent data handling across studies.

2D and 3D application-specific modules

Choose from a variety of assays—including 2D cell migration and invasion, immunotherapy potency assays, 3D spheroid/organoid analysis, wound healing, dilution cloning, virology applications, drug discovery, and cell-by-cell segmentation—using dedicated software modules that automate calculations and streamline reporting.

Regulatory-ready data management

Ensure data integrity with built-in 21 CFR Part 11 compliance features, including secure user authentication, full audit trails, electronic signatures, and controlled workflows—supporting both research and GMP environments.

Applications

Immune cell killing assays

Typical immune cell killing assays are labor-intensive and require users to collect multiple timepoints using a combination of techniques. In contrast, the label-free xCELLigence RTCA instruments provide a complete view of immune cell killing and closely mimics activity in vivo. Obtain highly sensitive, reproducible, and direct measurements of immune cell killing potency in real time. This simple and high throughput workflow also provides greater insight into cellular mechanism of action, serial killing, and exhaustion.

Viral cytopathic effect assays

Advance your anti-viral therapy or vaccine development by accurately measuring viral cytopathic effects (CPE) in real time with Agilent xCELLigence RTCA instruments, without the use of labor-intensive plaque assays. Monitor viral CPE automatically, from minutes to days, and greatly reduce workload and manual handling of samples. Measure cell proliferation kinetics, identify the optimal time point for viral infection with different cell seeding densities, and assess virus-mediated cytopathogenicity in one workflow.

Cytotoxicity assays

Determine cytotoxic effects of drugs or compounds by monitoring cell proliferation, cell size or morphology, and cell-substrate attachment quality in real time. Monitor the kinetics of long-term cellular responses with this simple and high-throughput cytotoxicity assay. Make informed decisions about the timing of cell treatments and easily generate dose-response curves at multiple time points, without increasing the workload.

Apoptosis and proliferation assays

Learn about apoptosis and proliferation assays that are simple, quantitative, and high throughput to provide multiple perspectives into cell health and viability. Measure real-time cell health directly and capture early and late apoptotic events such as modulation of cell adhesion, cell membrane integrity, and cell detachment. Track proliferation of host cells to detect readout for drug toxicity or virus presence. Detect and quantify apoptosis in living cells by label-free approach, or using apoptotic or nuclear labeling reagents. The xCELLigence RTCA eSight provides multiple temporal vantage points (impedance, fluorescence imaging, and brightfield imaging with automated AI-driven cell segmentation).
